چکیده

Background: Topical carbonic anhydrase inhibitors (CAI) are the commonly used pharmacologic agents as an ocular hypotensive in medical therapy of glaucoma. This traditional use is due to reduced aqueous production through decreased bicarbonate formation in ciliary body epithelium. Taken into account current economical burden worldwide, the well-known drugs are “rediscovered’ and repurposed. In this light of view CAI are highlighting new properties and new targets in modern ophthalmology. Objective: The objective of this review is to evaluate the evidence and discuss the rationale behind the recent suggestions that carbonic anhydrase inhibitors may be useful in therapy of retinitis pigmentosa.

Quantitative Structure - Activity Relationships Study of Carbonic Anhydrase Inhibitors Using Logistic Regression Model

Binary Logistic Regression (BLR) has been developed as non-linear models to establish quantitative structure- activity relationships (QSAR) between structural descriptors and biochemical activity of carbonic anhydrase inhibitors. Using a training set consisted of 21 compounds with known ki values, the model was trained and tested to solve two-class problems as active or inactive on the basi...

Therapeutic applications of glycosidic carbonic anhydrase inhibitors.

The zinc enzymes carbonic anhydrases (CAs, EC 4.2.1.1) are very efficient catalysts for the reversible hydration of carbon dioxide to bicarbonate and hence play an important physiological role. In humans, 16 different isozymes have been described, some of them being involved in various pathological disorders. Several of these isozymes are considered as drug targets, and the design of selective ...

Specificity and comparative activity of the carbonic anhydrase inhibitors neptazane and diamox on animal and human eyes.

IN an investigation of the function of the enzyme carbonic anhydrase in the formation of the aqueous humour (Langham, 1958b), studies were made with a new carbonic anhydrase inhibitor, 5-acetylimino-4-methyl-A2-1, 3, 4thiadiazoline-2-sulphonamide (Neptazane, Lederle Laboratories). The results of this study indicated that Neptazane was more active than the wellknown carbonic anhydrase inhibitor ...

Phosphate and Protein Concentrations of Intraocular Fluids. I. Effect of Carbonic Anhydrase Inhibition in Young and Old Rabbits.

The effects of a carbonic anhydrase inhibitor, dichlorphenamide, on the phosphate and protein concentrations of intraocular fluids have been studied. The administration of dichlorphenamide caused a marked increase in phosphate concentration of posterior and anterior aqueous humors. The changes in steady-state concentrations were compatible with a decrease in the flow coefficient of approximatel...
